http://hotelroomalerts.com/ a new service that alerts guests when their choice of hotel room opens up is here and we at hospemag.com think every hotelier should be on it.
First, what is it about?

POV - Hoteliers
You would have to create a waiting list and then follow up with the them when you had last minute cancellations or no-shows.
Boom - now it is automated.

Here is the official explanation of how it works:
How it Works
- Choose the hotel(s) and room(s) you are interested in.
- Enter an e-mail address and phone number (optional, for text-message notification) to be alerted of opening.
- Receive a text message and e-mail alert as soon as your room becomes available!
- Book it!
Availability Checking
We check hotels for availability 24/7 around-the-clock. By using our service, you will be the first to know when a booked hotel opens up!
Partial Availability
If your check-in period spans more than one day, we will e-mail you when we detect partial availability. Partial availability occurs when one or more of your desired rooms is open for only a portion of the dates you're interested in.
Partial availability also can occur when the entire duration of your stay is available through two or more different rooms.
It is not uncommon for a certain room to open up for one day, and a different room to open up for a different day. If you don't mind switching rooms within the same hotel during your check-in period, there is a greater chance to enable your stay. Partial availability is enabled by default.
